Liver sinusoidal endothelial cells (LSECs) are highly specialized cells that form the scavenger endothelium within the liver. They exhibit extraordinarily high uptake of soluble waste molecules (< 0.23 µm), thus playing an important role in blood clearance. The supply of primary LSECs is limited by the low and sporadic availability of human liver tissue. In addition, primary LSECs can only be kept for up to 1-2 population doublings in culture.
upcyte® LSECs were derived from primary LSECs from a single donor. These cells have an extended lifespan whilst maintaining primary cell characteristics. This means that large batches of upcyte® LSECs up to 2000 vials from a single donor with the same quality can be provided (cryo-vials with a minimum of 500.000 viable cells).
upcyte® LSECs have been tested for numerous cel type specific markers, scavenging / uptake functions, as well as the ability to form tubes in matrigel. In addition whole genome expression profiling revealed that upcyte® LSECs and primary LSECs have similar expression profile. Only 0.45% (218) of a total of 48,107 gene probes analyzed have been found to be up- or down-regulated more than 2-fold.
upcyte® LSECs exhibit a similar sensitivity to primary cells making them the ideal tool to study toxicity and to set-up co-culture models to generate a more predictive in vitro system. These cells can be used for screening to exclude compounds at an early stage meaning toxic compounds do not reach animal testing and clinical stages.

All cells have been tested for the absence of CMV, HIV, Hepatitis B and C and microbial contaminants (fungi, bacteria and mycoplasma). upcyte® cells are genetically modified cells (GMO, genetically modified organism) classified as risk group 1 and have to be handled in a genetic engineering facility carrying out genetic engineering work at Security Level 1 (e.g. S1 lab). upcyte® Hepatocytes and upcyte® Endothelial Cells are officially classified as risk group 1 organisms by the German Federal Office of Consumer Protection and Food Safety (BVL) and the Central Committee on Biological Safety (ZKBS).
